The Protective Effects of Sodium Pentaborate Tetrahydrate Against UVB-induced Apoptosis in Human Keratinocytes

چکیده
Ultraviolet radiation (UV) is an environmental carcinogen causing human skin cancer. Exposure of the to UV produces apoptotic keratinocytes called sunburn cells within epidermis. Boron, essential element for plants, has several biological properties, such as anti-cancer, anti-microbial, and anti-oxidant. In present study, possible protective effects sodium pentaborate pentahydrate (SPT) against UVB-induced apoptosis in keratinocyte cells, HaCaT, were investigated. They treated with SPT at different concentrations (7.8-125 μg/mL) 24h after UVB irradiation (20, 30 60mJ/cm2). Cell viability, annexin V assay, cell cycle analysis, apoptosis-related gene levels measured using RT-PCR. Treatment (15.6-31.25μg/mL) mJ/m2 exposure significantly increased survival. Annexin analysis demonstrated a robust effect by treatment 15.6 31.25μg/mL 30mJ/cm2 irradiation. The revealed that elevated number G0/G1 phase while G2/M phase, suggesting changes partially reversed. Furthermore, 15.6μg/mL blocked activation caspase 3, 9, Bax, p53. These results indicate exerts Thus, led strong protection death HaCaT cells.

UV-induced apoptosis in keratinocytes is a highly complex process in which various molecular pathways are involved. These include the extrinsic pathway via triggering of death receptors and the intrinsic pathway via DNA damage and reactive oxygen species (ROS) formation. In this study we investigated the effect of catalase and CuZn-superoxide dismutase (SOD) overexpression on apoptosis induced ...

متن کاملProtective effect of fermented Cyclopia intermedia against UVB-induced damage in HaCaT human keratinocytes
BACKGROUND The fermented leaves and stems of Cyclopia intermedia are used to brew honeybush tea, a herbal tea indigenous to South Africa. The aim of this study was to evaluate the protective effect of fermented honeybush extracts (FH ex) and scale-up fermented honeybush extracts (SFH ex) against ultraviolet B (UVB)-induced damage in HaCaT keratinocytes. To this end, we examined UVB-induced cell...
